Aging is frequenty associated with major factors responsible for functional limitation and motor dependency. Due to the demands upon the collagen-containing structures (movement and bearing weight), it is vital to optimize the conditions that support the maintenance of joint mobility and musculoskeletal condition.

In vivo study – protocol and results

This in vivo study was conducted by INSERM-I2MC (National medical Research institute, France) and lasted 14 weeks. It was carried on ovariectomized mice C57Bl/6 female mice (12 weeks old) having developed an osteoporosis. The objective was to evaluate the effects of an oral intake of Naticol® fish collagen peptides (daily diet containing 2,5% of Naticol®) on bone mineral density, muscular force and tissues. The mice received a normal chow diet.

Muscular resistance and strength

In order to assess muscular resistance and strength, we used the grip test. The mice groups treated with Naticol® showed a significant increase of their muscular force vs. control group (p<0,05).

Muscular fiber histological structure

Muscular fibers were analized by microscopy using hemalun/ eosin for muscle fiber structure and Masson trichrome for collagen. The mice group treated with Naticol® showed a significant increase of their muscular mass and consequently, a decrease of the deleterious matrix collagen (p<0,05).

This gain of muscular tissue may translate the higher muscular force we previously observed. In physiologyy, a muscle which gains muscular force builds myocytes.

Bone mineral density

Bone density was assessed by CT tomography. The results showed that bone density moved towards an increase in the mice group treated with Naticol® vs. control group.

Intensity of the signal (bones of the leg)

Conclusion

The results of oral ingestion of Naticol® on a daily basis, showed that regular intake of Naticol® Fish collagen peptides may improve the musculoskeletal condition.

In this study, Naticol® has demonstrated its role as a natural ingredient for musculoskeletal condition.

Rectocolitis is a chronic inflammation of the rectum and colon mucosa. It can be associated with diarrhea, abdominal pain, bloating and blood in the stool. More common in industrialized countries, rectocolitis is enhanced by genetic predisposition and immune system dysfunction. It is a condition characterized by alternating phases of activity (relapses) and phases free of symptoms (remission).

Naticol®Gut, patented fish collagen peptides developed by WEISHARDT, showed anti-inflammatory activities and effects on intestinal microbiota composition.

Protocol: rectocolitis study

In this study, female C57Bl/6 mice were used and treated with a chemical colitogen, dextran sulfate sodium (DSS), at a concentration of 1.5 % in the drinking water. Oral administration of DSS to the mice is widely used in models of chronic inflammatory bowel disease. DSS dissolved in drinking water is toxic to the epithelium overlying the colon of mice leading to a colitis characterized by weight loss and bloody diarrhea.

Naticol®Gut (patented fish collagen peptides developed by Weishardt) and casein were respectively added to the different mice groups treated with DSS.

RESULTS and DISCUSSION

The figure 1 shows that all mice treated with DSS 1,5 lost weight. Only the mice groups treated with DSS + Naticol®Gut, at the dosis of 0,1g Naticol®Gut/kg body weight/day recovered from weight loss contrary to DSS group and DSS + casein group.

The figure 2 shows the histological epithelial disruption present in both DSS group and DSS +casein group. No epithelial intestinal disruption was observed in the groups DSS +Naticol®Gut.

In the figure 3, macroscopic score of inflammation was significantly decreased with Naticol®Gut compared to DSS group (p <0,01). Macroscopic score is obtained by the sum of stool, colon damage, colon weight and colon length scores (Wallace score).

Cytokine expression and microbiota composition in colon

Naticol®GUT showed a decrease of pro-inflammatory cytokines levels, in particular a significant decrease of Il-6 and IL1-B levels. TGF-β was significantly increased which translates a repairing effect.

Regarding the intestinal microbiota composition (analysis by PCR), a statistically significant increase of Faecalibacterium praunitzii similar to control group (without colitis induced by DSS) was observed in the groups DSS+ Naticol®Gut. No significant increase was obtained in both DSS+ casein and DSS groups. Nota: *p<0.05 et **p<0.01 as compared to DSS.

Conclusion

Naticol®GUT are patented fish collagen peptides produced by WEISHARDT. In this study, they showed a decrease in the level of expression of pro-inflammatory cytokines (IL1-β, IL6 etc...) and an increase of antiinflammatory markers such as TGF-β. Here, treatment with Naticol®GUT demonstrated protective effects on the integrity of the epithelial tissue and positive effects on the intestinal microbiota composition, in particular on Faecalibacterium praunitzii.

Clinical study csp 3507 - methods and results

This clinical study was double-blind, randomized, against placebo. It was carried on 95 healthy female subjects, mean-aged 55,6 y.o, for 12 weeks. The objective was to evaluate the anti-aging potential of a daily oral intake of 2,5g of Naticol® fish collagen peptides on cutaneous measures and its tolerance. 2,5g of Naticol® powder was mixed with water and ingested in the morning. This study was conducted by CPCAD / Hospital of Nice (France) and biomechanical measurements of volunteers were controlled by certified dermatologists.

This clinical study was performed according to French regulatory competent authority (ANSM) and approved by the local Ethical Committee (CPP). Consent was obtained from each subject before entry in the study.

Clinical score of wrinkles

Clinical evaluation of facial wrinkles was performed by a trained clinician using Bazin-Doublet’s atlas.

The clinical assessment of the perioral and Crow’s feet wrinkles compared with baseline showed a significant decrease of the mean score of facial wrinkles at week 12.

At Week 12, an improvement in crow’s feet wrinkles was observed for more than 70% of the subjects in the 2.5g/d Naticol® treated groups. As well, about 60% of the subjects showed an improvement in perioral wrinkles in 2.5g/d Naticol® group.

Elasticity - Cutometer SEM 575

The Cutometer® DUAL probe (Courage and Kazhaka, Köln, Germany) using a suction method measured a deformation perpendicular to the skin surface. A negative pressure of 450 mbars was applied to the skin through the probe for 2 seconds, followed by a relaxation period of 2 seconds. This cycle was performed 5 times. This method measures the degree of deformation and the time required for the skin to return to its original state. The deformation induced on the skin is measured by an optical system.

A significant increase in skin firmness on the external part of the forearm was observed at Week 12 for 2,5g/d Naticol® while no significant changes over time were observed in the placebo group.

Skin radiance and skin complexion homogeneity

A visual analogic scale (VAS) was used by a trained clinician to assess the level of skin radiance between 0 (very dull skin) and 10 (very radiant skin). At the same manner, a visual analogic scale (VAS) was used to assess the level of skin complexion homogeneity between 0 (very heterogeneous complexion) and 10 (very homogeneous complexion).

Skin radiance and skin complexion homogeneity were statistically significantly increased for 2,5g/d Naticol® after Week 8 while no significant change was seen in the Placebo group.

Tolerance

It was evaluated through a clinical examination and volunteer’s questionnaire. Naticol® presents globally a good tolerance.

Conclusion

The results of oral ingestion of 2,5g Naticol® on a daily basis, up to 12 weeks, clinically showed that regular intake of Naticol® may improve skin elasticity and reduce facial wrinkle appearance. In this study, skin complexion homogeneity and radiance were also significantly improved after 8 weeks.

Clinical study csr3463 – methods and results

This clinical study was double-blind, randomized, against placebo and carried on 60 healthy female subjects (mean 56 y.o.). The objective was to evaluate the anti-aging potential of daily oral intake of 5g of Naticol® fish collagen peptides on cutaneous measures and its tolerance. 5g of Naticol® was mixed with liquid and ingested in the morning. This study was conducted by CPCAD (clinical pharmacology center specialized in dermatology - Hospital of Nice, France). All measurements (including biometry measurements) were controlled by certified dermatologists.

This clinical study was performed according to French regulatory competent authority (ANSM) and approved by the local Ethical Committee (CPP). Consent was obtained from each subject before entry in the study.

Skin relief – Visia® CR

Photographies of the face were taken using a standardized device, the VISIA® CR.

This device allows to take pictures using standardized lighting: white light, cross polarized light, parallel polarized light and UVA light. The replica was lit with incident light at 35° which produced shadowing behind each wrinkle.

Elasticity - Cutometer SEM 575

Cutometer® SEM 575 by COURAGE & KHAZAKA was used to measure a deformation perpendicular to the skin during 2 seconds, followed by a relaxation period of 2 seconds.

This method measures the degree of deformation and the time required for the skin to return to its original state. The deformation induced on the skin is measured by an optical system.

The measured parameters are: Ue, the elastic deformation; Ur, the elastic back deformation; Uf the total extensibility of the skin; Ua, the total deformation recovery; Ur/Ue the pure elasticity.

Tolerance

It was evaluated through a clinical examination and volunteer’s questionnaire. Naticol® presents globally a good tolerance.

Conclusion

In this clinical study, oral intake of 5g Naticol®/day, for 8 weeks, showed benefits on skin firmness and a reduction of wrinkle appearance. These results may also indicate a beneficial effect of Naticol®, fish collagen peptides, on the skin dehydration due to cold outside conditions.

Clinical study cp 3407 – methods and results

This clinical study was double-blind, randomized, against placebo, done on two groups and was carried on 40 healthy female subjects from 50 to 70 y.o, during spring and for 12 weeks. The objective was to evaluate the anti-aging potential of oral intake of 10g of Naticol® fish collagen peptides on cutaneous measures and its tolerance. 10g of Naticol® powder was mixed with liquid and ingested in the morning. This study was conducted by CPCAD (independent pharmacology center specialized in dermatology from Nice, France) and biomechanical measurements of volunteers were controlled by certified dermatologists.

This clinical study was performed according to French regulatory competent authority (ANSM) and approved by the local Ethical Committee (CPP). Consent was obtained from each subject before entry in the study.

Hydration - Corneometer CM825

This device measures the electrical capacitance of the skin which is expressed as an index of hydration. It allows to assess the hydration state of the first layers using electrical capacitance measurements.

The perception of the volunteers about how Naticol® performs was a main component of the clinical study. All the women completed self-assessment questionnaires at the onset of the trial and at the end of the study.

It was evaluated through a clinical examination and volunteer’s questionnaire. Naticol® presents globally an excellent tolerance.

Conclusion

The results of oral ingestion of 10g Naticol® on a daily basis, up to 12 weeks, clinically show that regular intake of Naticol® Fish collagen peptides should improve the basic skin moisture and elasticity, and reduces wrinkles.
